Chromatography may be defined as, “an analytical technique for identification and separation of various components of a mixture on the basis of difference in their affinity for stationary and mobile phases”.
The mixture is applied to either a solid or liquid phase called stationary phase
Liquid or gas phase is allowed to move slowly over the stationary phase , called mobile phase.
Types of chromatography:
Chromatographic process may be classified as
1. Adsorption chromatography
2. Partition chromatography.
3. Ion- exchange chromatography.
Advantages:
Large volume of samples can be separated.
Separation of a mixture of similar molecules is possible only in adsorption in adsorption chromatography.
Applications:
Purification of substances from their contaminants
Identification of products
